A Rapid Method for Directed Gene Knockout in Cyprinus carpio var. wuyuanensis

Hebao red carp (Cyprinus carpio var. Wuyuanensis) is an allotetraploid cyprinidae fish, which has undergone the unique fourth round of full genome replication of cyprinidae fish. It has good cross affinity and can cross with a variety of carp and crucian carp. The progeny has obvious heterosis, which plays an important role in promoting the aquaculture industry in China. Compared with traditional fish breeding techniques, CRASPR/Cas9 gene editing technology has a stronger purpose, shorter time and lower cost in breeding new varieties. It can achieve accurate editing of target genes and quickly obtain homozygous mutants of target traits, which has a certain prospect for improving fish breeding efficiency and germplasm resources. We identificatied our target gene in the genome of Hebao red carp and simutaneously first established the CRISPR/Cas9 editing system in Hebao red carp. Finally we obtained F0 mutants, which provides research basis for the breeding development of Hebao red carp and optimization of breeding traits.
